【摘要】 目的 探讨白细胞介素 １３（ ＩＬ－１３）对体外培养的大鼠系膜细胞的增殖及其产生白细胞介素１β（ＩＬ－１β）的影响。方法 用四甲基偶氮唑（ＭＴＴ）法测定系膜细胞增殖，用逆转录聚合酶链反应（ＫＴ－ＰＣＲ） 及酶联免疫吸附（ＥＬＩＳＡ）法测定系膜细胞ＩＬ－１β ｍＲＮＡ表达及ＩＬ－１β蛋白水平。结果ＩＬ－１３在 １．０、１０、１００ ｎｇ／ｍｌ浓度范围呈剂量依赖性地抑制系膜细胞的增殖；含 ５％小牛血清（ＦＣＳ）的ＲＰＭＩ １６４０培养状态下的系膜细胞检测不出 ＩＬ－１βｍＲＮＡ的表达，加入脂多糖（ＬＰＳ）可诱导系膜细胞表达ＩＬ－１βｍＲＮＡ及分泌ＩＬ－１β，ＩＬ－１３在抑制系膜细胞的增殖的相应浓度范围可显著地抑制ＬＰＳ诱导的系膜细胞ＩＬ－１β ｍＲＮＡ表达及 ＩＬ－１β分泌。ＩＬ－１３在 １０ ｎｇ／ｍｌ浓度时即可几乎完全抑制 ＬＰＳ诱导的系膜细胞 ＩＬ－１β ｍＲＮＡ表达。结论 ＩＬ－１３对体外培养的系膜细胞增殖及炎症效应具有抑制作用。更多还原

【Abstract】 Objective To explore the effects of interleukin-13(IL-13) on cell proliferation and interleukin-113(IL-1β) production in mesangial cell. Methods Cell proliferation was tested by MTT method. The expression of IL-1β mRNA in mesangial cells was evaluated by RT-PCR. The protein synthesis of IL-1β in mesangial cells was measured by ELISA. Results IL-13. in the range of concentration from 1 ng/ml to 100 ng/ml. could inhibit the proliferation of mesangial cells with or without inducing by LPS: Il-13. in the same range of concentration, also inhibited obviously the expression of IL-1β mRNA and the protein release of IL-1β induced by LPS in mesangial cells. IL-13 in 10 ng/ml could inhibit almost completely the expression of IL-1β mRNA induced by LBS. Conclusion IL-13 can inhibit the proliferation of mesangial cells and IL-1βproduction induced by LBS in mesangial cells.更多还原
