【摘要】 目的　基质金属蛋白酶 2 (MMP 2 )mRNA和蛋白在卵巢上皮性肿瘤组织中的表达以及与临床病理特征和预后的关系。方法　用半定量逆转录聚合酶链反应技术、链霉菌抗生物素蛋白 过氧化物酶连接法对 41例卵巢上皮性癌及 2 6例卵巢良性肿瘤组织中MMP 2mRNA及蛋白的表达分别进行定量和定位分析 ;应用蛋白印迹法定性检测MMP 2酶原和活性MMP 2在卵巢上皮性肿瘤组织中的表达。结果　良、恶性卵巢肿瘤组织中MMP 2mRNA表达分别为 0 6 8± 0 48和 0 97± 1 0 0 ,MMP 2蛋白表达阳性率分别为 5 4%和 76 % ,两者分别比较 ,差异无显著性 (P >0 0 5 )。卵巢上皮性癌组织中的活性MMP 2表达阳性率为 71% ,显著高于卵巢良性肿瘤组织的 42 % (P <0 0 5 )。手术病理分期为Ⅲ～Ⅳ期卵巢癌患者的活性MMP 2表达阳性率为 81% ,显著高于Ⅰ～Ⅱ期患者的 9例中 3例 (P=0 0 1)。病情进展患者的活性MMP 2表达阳性率为 86 % ,明显高于无进展患者的 19例中 10例 (P=0 0 2 )。结论　卵巢上皮性肿瘤普遍存在MMP 2mRNA及蛋白表达 ,活性MMP 2表达与卵巢上皮性癌侵袭、转移密切相关 ,是卵巢上皮性癌的一项预后监测指标更多还原

【Abstract】 Objective To investigate the relationship between the expression of matrix metalloproteinase 2 (MMP 2) and clinical characteristics of patients with epithelial ovarian tumor Methods Reverse transcriptase ploymerase chain reation and immunohisto chemistry were used to detect the expression of MMP 2 in 41 epithelial ovarian cancer and 26 ovarian benign tumors Western blot analysis was used to examine proMMP 2 and active MMP 2 in these tumors Results The expression of MMP 2 gene and protein was not significantly higher in malignant tissues than that in benign ones The positive percentage of active MMP 2 expression in epithelial ovarian cancer was 71% and 42% in ovarian benign tumor ( P <0 05) The positive percentage of active MMP 2 in stage Ⅲ and Ⅳ epithelial ovarian cancer was significantly higher than in stageⅠ and Ⅱ epithelial ovarian cancer ( P <0 05) The positive percentage of active MMP 2 in progressive patients with epithelial ovarian cancer was significantly higher than in patients in remission ( P <0 05) Conclusions MMP 2 gene and protein were generally expressed in epithelial ovarian tumor There was a significant correlation between active MMP 2 and the invasion and metastasis of epithelial ovarian cancer Active MMP 2 is a prognostic indicator in patients with epithelial ovarian cancer更多还原
