神经生长因子对实验性自身免疫性脑脊髓炎的保护作用
Suppression of Experimental Autoimmune Encephalomyelitis by Intramuscular Administration of Human Nerve Growth Factor
【摘要】 目的 建立实验性自身免疫性脑脊髓炎 (EAE)动物模型 ,探讨神经生长因子 (h NGF)对豚鼠 EAE的保护作用。方法 豚鼠经一次足跖真皮内注射完全福氏佐剂 -豚鼠全脊髓匀浆 ,诱发 EAE疾病 ;在豚鼠致炎前注射 h NGF,观察其对 EAE临床症状、发病率及脊髓病变的影响。结果 模型组豚鼠免疫注射后相继出现明显的 EAE症状 ,发病率为 9/1 0。豚鼠在致炎前注射 h NGF,可明显控制病情 ,降低 EAE发病率 ,明显改善脊髓的病理改变 ,且 h NGF的作用呈剂量依赖性。结论 h NGF对实验性自身免疫性脑脊髓炎有显著的保护作用
【Abstract】 AIM The purpose is to observe the effect of human nerve growt h factor(hNGF) on experimental autoimmune encephalomyelitis(EAE)in guinea pig. METHODS EAE model was produced in guinea pig by intradermally injecting CFA-GPS CH prepared with guinea pig spinal cord homogenate in complete Freund′s adjuvan t . Guinea pigs were divided into 4 groups. In group 1, 2 and 4, 200 IU/kg, 400 IU / kg of hNGF and normal saline were intramuscularly administrated before sensitiza tion respectively; and in group 3, normal saline was intramuscularly administrat ed to normal animals. The clinical symptoms and incidence of EAE were observed f o r 30 d after sensitization, after that the spinal cord was removed for routine histological test. RESULTS In model group, the animal of 9/10 dev eloped moderate to severe clinical symptom of EAE. hNGF im before sensitization significantly ameliorated the symptom and histopathology of spinal cord in dose -dependent manner and reduced inciden ce of EAE. CONCLUSION The results indicate that the hNGF has a pr otective effect on CFA-GPSCH-induced EAE in guinea pig.
