生物素标记的6种cDNA探针检测S180及其克隆细胞株mRNA的表达
The Difference of mRNA Expression in S180 Cell Strain and Its Cloned Cells
【摘要】 目的 检测S180及其克隆细胞株S1B11及S2D9mRNA的表达 ,对这些细胞株进行识别和质量控制。方法 用生物素标记的 6种cDNA探针 ,细胞玻片原位杂交的方法检测细胞中mRNA的表达。结果 北京市肿瘤研究所 (肿瘤所 )保存的S180与生物素标记的P16、c fos、c myc及c jun探针杂交阳性 ,克隆细胞株S1B11与c fos及c jun探针杂交阳性 ,克隆细胞株S2D9与c fos、c myc及c jun探针杂交阳性。结论 肿瘤所S180及其 2株克隆细胞中mRNA的表达不同 ,c myc基因的表达与否可以把S1B11及S2D9克隆细胞区别开 ;细胞株致瘤性与癌基因表达有关
【Abstract】 Objective To examine the difference of mRNA expression in S180 cell strain and its different cloned cells.Methods The probes labeled by biotin were used in situ hybridization to detect the expression of 6 kinds of mRNA that were p16, c fos, c jun, c myc, p21 and p53.Results P16,c fos, c jun and c myc mRNAs were found in Cancer Institute S180 cells, c fos and c jun mRNAs in S1B11 cloned cells, c fos, c myc and c jun mRNAs in S2D9 cloned cells. Conclusion The expression of mRNAs was different in Cancer Institute S180 and 2 different cloned cell strains and c myc probe could distinguish S1B11 from S2D9 cloned cells; expression of mRNAs was associated with carcinogenesis.
