鸡腺胃型传染性支气管炎病毒金坛分离株核蛋白基因的克隆和测序

【摘要】 应用逆转录多聚酶链反应技术，参照 Ｂｏｕｒｓｎｅｌｌ等发表的鸡传染性支气管炎病毒核蛋白基因序列，自行设计并合成１对引物，应用该引物，从感染ＩＢＶ金坛株的第五代ＳＰＦ鸡胚尿囊液中扩增出片段大小为８２８ｈｐ的ＪＴ株 Ｎ基因，与设计相符，表明扩增片段为 ＩＢＶ ＪＴ株 Ｎ基因。将扩增片段与 ｐＧＥＭ－ Ｔ Ｅａｓｙ载体连接，经核酸序列测定，与Ｇｅｎｅｂａｎｋ中报道的ＩＢＶ其它株Ｎ基因同源性较高。

【Abstract】 One pair of primers for amplifying the nucleocapsid gene of avian infectious bronchitis virus(IBV) had been designed. The amplified fragment was 828 hp from JT strain by RT - PCR. The N gene of JT strain was inserted into pGEM - T Easy vector and identified by sequencing. The homologies of nucleie acids between, JT strain and other strains is very high.
