魔芋ADP-葡萄糖焦磷酸化酶大亚基cDNA片段的克隆

【摘要】 利用ADP -葡萄糖焦磷酸化酶的保守序列设计引物 ,采用RT PCR方法 ,从白魔芋球茎组织的总RNA中扩增出目标cDNA片段 ,并克隆到pUC18载体中。序列分析后确认该片段属于一新的ADP -葡萄糖焦磷酸化酶大亚基基因

【Abstract】 PCR primers were designed based on the consensus domain of ADP glucose pyrophosphorylase.A cDNA fragment was amplified from the total RNA isolated from the yang corm of Amorphophallus albus via RT PCT and cloned into plasmid pUC18.The determined sequence showed this cDNA fragment was homologous to the large subunit gene of ADP glucose pyrophosphorylase.
