铈离子及其配合物对寡聚脱氧核苷酸的水解断裂作用
Oligodeoxynucleotide Hydrolysis by Cerium Ion and Its Coordinate Compound
【摘要】 报道了铈离子及其配合物对人工设计合成的 2 6个碱基的单链寡聚脱氧核糖核酸 (ODN)的水解断裂作用。Ce3+ 需要有氧气存在的条件下 ,氧化生成Ce4 + 后才能切断ODN。研究了各种条件下Ce4 + 与ODN的切断反应 ,在酸性和弱碱性条件下都能水解切断ODN ,在近中性时切断作用最强。随着反应温度的升高 ,Ce4 + 浓度的增大 ,反应时间的延长 ,切断反应越来越明显。Ce4 + 与氮三乙酸生成配合物以后也能切断ODN。Ce4 + 可作为切断ODN的分子剪刀。
【Abstract】 Cerium ion hydrolysis of ODN which has the sequence of 5′>GGAGGCGAACGATACGCA TCGACTGT<3′ was studied. Oxygen is required in the reaction of Ce 3+ scission ODN, because Ce 3+ is being oxidated to Ce 4+ at first and the resultant Ce 4+ is responsible for the DNA hydrolysis. The reaction of Ce 4+ with ODN was studied in various conditions. In the experiments, Ce 4+ cleavage the phosphodiester linkage both at acid and alkalinity conditions, while the cleavage reaction is most strongly near neutral (pH 7 3). The hydrolysis will be more and more obvious at higher temperature, higher concentration of Ce 4+ and longer reaction time. ODN can also be scissioned by coordinate compound formed from Ce 4+ and NTA, but this reaction is weaker than the reaction of Ce 4+ with ODN. The denaturing ploy acrylamide gel electrophoresis silver dye method was used to analyze the hydrolysis product.
