【摘要】 目的观察大蒜素 (allitridi,Alt)及葛花露 (Puerariaeflosessence ,PFE)对扑热息痛 (Paracetamol,Par)所致小鼠肝损伤的保护作用。方法建立小鼠Par肝损伤模型 ,分别测定生理盐水对照组、Par模型组、Par+Alt1 0mg/kg组、Par +Alt组 ( 1 0、2 0和 30mg/kg)、Par +PEE组小鼠血清天冬氨酸转氨酶(AST)、谷胱甘肽S -转移酶 (GST)活性及肝组织中谷胱甘肽 (GSH)和MDA含量。结果Alt可阻抑Par小鼠血清AST、GST和肝组织MDA升高及GSH耗竭 (均P <0 .0 1 ) ,其中抑制血清GST升高和肝内GSH耗竭的作用皆呈现量效关系 ;PFE亦可显著阻止Par小鼠血清AST、GST和肝组织MDA升高 ,对肝脏GSH耗竭则无保护作用 (P <0 .0 5 )。结论Alt小剂量即可有效防治Par肝损害 ,适当增加剂量更有利于Par鼠肝功能全面恢复 ;PFE对Par肝损害亦有显著保护作用 ,但不及Alt。更多还原

【Abstract】 Objective To observe the protective effects of allitridi(Alt) and puerariae flos essence(PFE) on murine hepatosis induced by paracetamol (acetaminophen,Par or APAP).Method The liver injury model induced by paracetamol was established in mice.The animals were divided in four groups which respectively treated by normal saline(control),paracetamol(Par),Par+Alt(according to dosage redivided in 3 subgroups-10,20 and 30mg/kg) and Par+PFE.Then,the activity of asparate aminotransferase(AST) and glutathione s transferase (GST) in serum as well as the contents of MDA and glutathione (GSH) in liver homogenate were tested in mice of all groups.Results Alt obviously inhibited either the increment of serum AST,GST and the hepatic MDA content or the exhaustion of hepatic GSH in Par treated mice (all P<0.01).Its inhibition effect on serum GST activity and hepatic GSH descent were dose depended strongly.PFE can also inhibit the increment of serum AST?GST and hepatic MDA,but showed no protective effect on exhaustion of murine hepatic GSH induced by Paracetamol.Conclusion The low dosage of ALT(10mg/kg) can protect murine liver from being injuried by Par.And proper increment of dose of Alt can foster the recovery of murine hepatic function in all the aspect in Par treated mice.PFE can also produce protective effect on murine Par induced hepatosis,which may be inferior to Alt.更多还原
