【摘要】 目的 :观察脑脊液白血病淋巴细胞及鞘注治疗前后细胞内DNA、RNA相对含量的变化。方法 :采用激光扫描共聚焦显微术 (LSCM)对 15例急性淋巴细胞性白血病患者脑脊液中 5 5 0个白血病淋巴细胞和鞘注治疗后脑脊液中 380个白血病淋巴细胞内DNA、RNA相对含量进行定量检测 ,同时用荧光强度地形分布图像表示 ,并与正常脑脊液中小淋巴细胞内DNA、RNA相对含量进行对比。结果 :脑脊液白血病淋巴细胞内DNA、RNA相对含量显著高于正常对照 (P <0 0 1) ;鞘注治疗后细胞内DNA、RNA相对含量明显低于治疗前 (P <0 0 1) 。且三者DNA、RNA荧光强度地形分布立体图像也存在着显著的差别 :治疗前似圆柱形 ,其顶部显强白色荧光 ,连成一片、完整无损 ;治疗后呈锯齿状 ,顶部显白色荧光 ,较治疗前弱 ,山峰较多 ,分散、断连 ;对照组似圆锥形 ,顶部显白色荧光。结论 :利用LSCM对脑脊液白血病淋巴细胞内DNA、RNA相对含量的定量分析可为CNLL诊断、鉴别诊断以及化疗药物的筛选、疗效评价、预后判断提供可靠的实验室依据更多还原

【Abstract】 Objective:To observe the change of DNA and RNA contents of luekemic lymphocyte in cerebrospinal fluid(CSF)prior to and following intrathecal chemotherapy in patients with acute lymphoblastic leukemia(ALL).Methods:DNA and RNA relative contents of 550 leukemic lymphcyets before and 380 lymphocytes after intrathecal chemotherapy were measured quantitatively by means of laser scanning confocal microtechnic(LSCM) in CSF of 15 patients with ALL,and 50 small lymphocytes from 25 cases’ normal CSF were used as control.Results:DNA and RNA contents of CSF leukemic cells in ALL were significantly higher than that of normal subjects (P<0 001),and the contents after intrathecal chemotherapy were significantly lower than before.The fluorescent intensive topography map of CSF leukemic lymphocyte showed cylinder in shape.It was quite different from the circular cone showed by normal small lymphocyte.After intrathecal chemotherapy,it appeared weaker fluorescence,like mountain peak.Conclusion:It could offer reliable laboratory foundation for early diagnosis,differential diagnosis,treatment and prognosis criterion of central nervous system lymphocyte leukemia(CNLL) to quantificationally analyse DNA and RNA contents of CSF Luekemic lymphoeytes in ALL by means of LSCM.更多还原
