高对映选择性环氧化物水解酶产生菌的筛选及特性研究

【摘要】 从土壤中分离的芽孢杆菌BacillusmegateriumECU1 0 0 1所产环氧化物水解酶能高对映选择性水解缩水甘油苯基醚 (对映选择率E值可达 47 8) ,当转化率为 5 5 9%时 ,剩余的 (S) 缩水甘油苯基醚的光学纯度 (对映体过量值 ,ee)可达 99 5 % ;当底物浓度提高到 60mmol/L时 ,光学纯 (S) 缩水甘油苯基醚的收率达到 2 5 6%

【Abstract】 A bacterial strain (ECU1001) was isolated from soil and identified to be Bacillus megaterium.The strain shows an activity of epoxide hydrolase that catalyzes the asymmetric hydrolysis of phenyl glycidyl ether,yielding (S)-epoxide and (R)-diol with high enantioselectivity (E=47.8).When the racemic substrate was enzymaticlly hydrolyzed to 55.9% conversion,the enantiomeric excess (ee) of recovered (S)-epoxide was 99.5%.The yield of (S)-phenyl glycidyl ether could reach 25.6% when concentration of the substrate was increased to 60 mmol/L.
