K562细胞对c-myc反义寡核苷酸的摄入及分布
Cellular uptake and distribution of c-myc antisense oliognucleotides in K562 cells
【摘要】 目的动态观察白血病细胞K562对经不同化学修饰的c-myc反义寡核苷酸(ASONS)及其脂质体复合物的摄入及在细胞内分布情况,以探讨与其生物学作用的关系。方法应用激光扫描共聚焦技术观察不同时间点K562细胞对上述物质的摄入量及分布特点。结果K562细胞对经脂质体包裹的c-myc ASONS及未包裹全硫化ASON的摄入量随培养时间的延长而增加,至5h达高峰,7h已明显下降。未包裹的ASONS摄入量大小为:全硫化组>端点硫化组>未硫化组,其荧光主要以点状、团块状形式分布在细胞表面及胞浆内,而经阳离子脂质体包裹的ASONS的荧光向核内聚集。结论对 ASONS进行硫化修饰可以促进 K562细胞对 c-myc A-SONS的摄入,脂质体作为载体可使ASONS向核内分布,从而可能影响ASONS生物学作用的发挥。
【Abstract】 Objective To observe the cellular uptake and their intracellular distribution of c-myc antisense oligonucleotides(A- SONS) and oligo-liposome complexes in K562 cells dynamically,in order to investigate the association with their biological effects. Methods Laser acanning confocal techanique was used to observe the conditions of uptake and localization of the above substances in K562 cells on the different time spots. Results Treated by oligo-liposome complexes or S-oligo,cellular uptake was significantly in- creased along with prolonged culture time.Cultured for 5h,cellular uptake arrived at the peak;it dropped obviously for 7h.In oligogroup,the comparison of uptake in three ASONS was:all phosphorothicate group>three-prime phosphorothioate group>unmodified group. And their fluorenscence distribution was on the cell surface or in the cytoplasm in punctation or mass pattem;while complexed with cationic liposome, the fluorenscence was located mainly in the nucleus. Conclusion Phosphorothioate of ASONS may increase the cellular uptake of c-myc ASONS and CATIONIC LIPOSOME may lead to THE accumulation of ASONS in nucleus of K562. This characteristics may influence their biological effects.
