p21WAF1和pRb在膀胱移行细胞癌中的表达及其意义
Expression of p21WAF1 and pRb in bladder transitional cell carcinoma
【摘要】 目的 探讨p2 1WAF1和 pRb在膀胱移行细胞癌 (BTCC)中表达及相互关系和其意义。 方法 应用免疫组织化学SP法检测 5 7例BTCC患者癌组织中P2 1WAF1和PRb的蛋白表达。结果 p2 1WAF1和 pRb的阳性表达率分别为 3 6 8%和4 5 6% ,p2 1WAF1随病理分级升高阳性率显著下降 (P <0 0 5 ) ,pRb的表达与BTCC的临床分期和有无转移均相关 ;p2 1WAF1和pRb的表达呈负相关 (P <0 0 0 5 ,Kappa =- 0 .4 0 1) ,p2 1WAF1( - ) /pRb( + )组合的发生率为 3 8 6% ,并与BTCC的病理分级、临床分期和有无转移均相关 (P <0 0 5 )。结论 p2 1WAF1和 pBb的表达异常与BTCC的发生发展密切相关 ,p2 1WAF1的改变可能为癌变的早期事件 ,联合检测 p2 1WAF1和 pRb可较完整准确地评价BTCC的生物学特性 ,估计预后 ,指导治疗。
【Abstract】 Objective To investigate the expression and significance of p21WAF1 and pRb in human bladder transitional cell carcinoma(BTCC),and the relationships between the two proteins. Method Immunohistochemistry was applied to detect the expression of p21WAF1 and pRb in 57 cases of BTCC. Results Postive rates of p21WAF1 and pRb in 57 cases of BTCC were 36.8% and 45 6%, respectively. p21WAF1 gene expression was related to the tumor grade ( P <0 005),pRb gene expression was related to the clinical stage and metastasis ( P <0 005),and p21WAF1 gene expression was negatively related to pRb gene expression ( P <0 005,Kappa=-0 401). Rate of p21WAF1(-)/pRb(+)gene expression was 38 6%,relating to tumor grade, clinical stage and metastasis simutaneously( P <0 05). Conclusion Alteration in at least one of the two genes might be required for most BTCC occuerence and develepment. Loss of the p21WAF1 protein may be an early event in tumorigenesis. The analysis of the two genes is more valuable for evaluating the biological character of BTCC.
