以壳聚糖为载体金属螯合亲和层析法纯化猪红细胞Cu·Zn-SOD

【摘要】 从虾壳制备所得壳聚糖为基质合成金属螯合亲和吸附剂 ,用于纯化猪血铜锌超氧化物歧化酶 (Cu·Zn -SOD) ,得到比活为 4 75 6U·mg- 1的酶蛋白 ,收率为 67% ,纯化倍数为 6 1。聚丙烯酰胺凝胶电泳及活性染色定位表明 ,经螯合层析纯化后的酶纯度基本均一
【Abstract】 Cu·Zn-SOD from porcine red cells was purified by metal chelate affinity chromatography(MCAC)using chitosan prepared from shrimp shell as substrate.The SOD was purified to homogeneous in one step affinity chromatography.Its specific activity is 4756 U·mg -1 and recovery is 67%.
