慢性心肌冬眠动物模型的建立

【摘要】 目的 :建立慢性心肌冬眠动物模型。方法 :随机选择健康杂种犬 2 2只 ,使冠状动脉左前降支缩窄 5 0 % ,继续喂养 1个月以上 ,行第二次手术 ,暴露犬左心室和左前降支狭窄处 ,进行 :①多巴酚丁胺超声心动图负荷试验 ;②潘生丁负荷试验 ;③心肌声学造影 ;④氯化三苯基四氮唑 (TTC)染色 ;⑤病理学检查。结果 :共 10只犬完成了所有实验步骤 ,存活时间 2 5~ 5 5d ,平均 (4 5 .2± 9.5 9)d。①结扎后与结扎前冠脉内径比为 :0 .4 4~ 0 .5 4,平均 0 .4 8± 0 .0 4 ;② 10只犬均出现并保持了节段性室壁运动异常 ;③多巴酚丁胺负荷试验中 ,所有犬均出现室壁运动改善及“双相反应” ;④潘生丁负荷试验表明冠脉血流储备明显降低 ;⑤运动异常节段声学造影后的灰阶强度较正常节段明显减低 ;⑥术后TTC染色及光镜和电镜证实 3只犬出现小灶性透壁性心肌坏死 ,1只犬出现小灶性心内膜下心肌坏死 ,余 6只犬均无心肌坏死发现。结论 :在动物实验中 ,建立慢性 (>1mo)心肌冬眠的动物模型是可行的。

【Abstract】 Objective:To establish the animal model of chronic myocardial hibernation.Methods:A left anterior descending coronary artery (CA) stenosis was created and maintained for 25~55 survived days ( mean of 45.2±9.59 days) (n=10) to reduced the CA diameter by a mean of 50% with severe regional myocardial systolic dysfunction. Dobutamine stress echocardiography,Dipyridomole stress test,myocardial contrast echocardiography,TTC stain,and pathomorphology examination were performed.Results:①The CA diameter after and before ligation were (0.84±0.14)mm and (1.72±0.19)mm respectively,and their the ratio was 0.44~0.54 (mean of 0.48±0.04);②Dobutamine stress test showed that the biphasic response was reserved in all 10 dogs;③the velocity of the stenotic CA increased significantly(P<0.05) after using dipyridomole, and the coronary flow reserve decreased significantly; ④Compared to the segments with normal movement, the myocardial visual density in segments with abnormal movement were decreased significantly;⑤TTC stain showed that there were localized myocardial necrosis in 4 dogs, and there were no myocardial necrosis in rest 6 dogs. The results were consistent with those found by microscope and electroscope.Conclusion:The establishment of the animal model of chronic myocardial hibernation is feasible.
