维甲酸镍配合物的合成及其抑制白血病K562细胞增殖的研究
Studies on Synthesis and Inhibitory Effect of Nickel Complex with Retinoic Acid on Proliferation of Leukemia K562 Cell
【摘要】 为了开发一种新型高效的维甲类抗癌新药 ,在无水、无氧、避光条件下合成了维甲酸镍的配合物 ,通过元素分析、摩尔电导、红外光谱、紫外光谱、差热及核磁共振氢谱测定了配合物的组成、结构和性质 ,其分子式为 Ni(RA) 2 · 3H2 O.采用 MTT比色法 ,克隆形成法研究了该配合物及全反式维甲酸 (ATRA)对 K56 2细胞增殖的影响 .结果表明 ,二者均具有抑制 K56 2细胞增殖的作用 ,尤其该配合物能强烈抑制 K56 2细胞的增殖 ,作用明显优于全反式维甲酸 (p <0 .0 0 1)
【Abstract】 In order to develop a highly effective antitumor drug of retinoic acid,a new complex of nickel with all trans retinoic acid has been synthesized in absolute ethanol,without oxygen in darkness and characterized by elemental analysis,molar conductance,IR,UV,and HNMR spectrometry. Its formula is best described as Ni(RA) 2 ·3H 2O. The inhibitory effects of ATRA and its nickel complexes on K562 cell lines have been investigated by using MTT colorimetric assay and clonic assay. The result showed that ATRA and its nickel complexes Ni(RA) 2 ·3H 2O both have inhibitory effects on K562 cells. Especially,the complex Ni(RA) 2 ·3H 2O has a far stronger inhibitory effect on K562 cells than the ligand ATRA (p<0.001).
