SLE患者淋巴细胞核基质蛋白的变化及临床意义

【摘要】 目的 :比较SLE患者与正常人及SLE患者静止期与活动期淋巴细胞核基质蛋白的变化 ,观察活动期SLE患者特有的淋巴细胞核基质蛋白对SLE患者骨髓CFU -GM生成的影响。方法 :用高盐法提取核基质蛋白 ,SDS -PAGE电泳比较核基质蛋白的差异 ,体外甲基纤维素法培养骨髓CFU -GM。结果 :SLE患者的淋巴细胞核基质在分子量为 2 5kd、45kd、75kd、80kd处出现正常人所没有的蛋白带 ,分子量为 40kd、70kd的核基质蛋白为活动期SLE患者淋巴细胞特有 ,培养体系中加入 70kd核基质蛋白的SLE患者骨髓CFU -GM产率为 38.2 1± 18.70 ,显著低于对照组 (P <0 .0 0 1)。结论 :SLE患者的淋巴细胞核基质蛋白与正常人相比有明显差异 ,分子量为 2 5kd、45kd、75kd、80kd的核基质蛋白可能与SLE的发生发展有关 ,分子量为70kd的核基质蛋白抑制SLE患者骨髓CFU -GM的生成 ,可能与SLE的活动有关。

【Abstract】 Objective: To compare the composition of nuclear matrix proteins(NMPs) between SLE and control and between active and static of SLE. To observe the influence of active NMPs on SLE bone marrow CFU-GM. Methods: We extracted NMPs with high-salt, compared difference of NMPs with one dimensional denaturing gel electrophoresis (SDS-PAGE), cultured CFU-GM of SLE bone marrow in vitro with semisolid methylcellulose. Results: New populations of NMPs were observed in SLE, which were 25Kd?45kd?75kd?80kd.NMPs of 40kd?70kd appeared in active SLE, specifically. The product of CFU-GM was 38.21±18.70 in the presence of 70kd NMPs, which was lower than that in control ( P <0.001). Conclusion: There is significant difference between SLE and control. NMPs of 25kd?45kd?75kd?80kd may related to pathogenesis of SLE. NMPs of 70kd in active SLE suppress bone marrow CFU-GM producing of SLE, it may related to activation of SLE.
