【摘要】 目的 :分析不明原因感音神经性聋 (ISNHL )患者血浆中热休克蛋白 (HSPs)抗体的成分 ,探讨 HSPs抗体存在的意义。方法 :以纯化的人类重组 H SP2 7、HSP6 0、HSP71、HSP90 α和 HSP90 β为抗原 ,用免疫转印迹技术检测 ISNHL、系统性红斑狼疮及正常健康者血浆中相应的抗体。结果 :ISNH L 组血浆中 H SP2 7、HSP6 0、H SP71、HSP90 α和 HSP90 β抗体阳性率分别为 12 .1%、7.3%、5 1.2 %、4.8%、0 % ,其中 HSP71抗体阳性率显著高于系统性红斑狼疮组和正常对照组 (P <0 .0 1)。结论 :HSP71抗体是 ISNHL 患者体内主要的热休克蛋白抗体 ,该抗体的存在预示某些 ISNHL 可能与自身免疫有关。

【Abstract】 Objective:To identify the specificity of autoantibodies to heat shock proteins in idiopathic sensorineural hearing loss (ISNHL) and to evaluate the existence of autoantibodies.Method:Plasma samples were obtained from patients with ISNHL (n=41), systemic lupus erythematosus (n=22), and normal controls (n=30). Plasma autoantibodies to heat shock proteins (HSPs) were detected by Western blot using purified recombinant human HSP27?HSP60?HSP71?HSP90 α and HSP90 β as antigen respectively.Result:In the ISNHL, the incidence of plasma autoantibodies to recombinant human HSP27? HSP60?HSP71?HSP90 α and HSP90 β were 12.1 %? 7.3 %? 51.2 % ? 4.8 % and 0% respectively. The incidence of autoantibody to HSP71 was significantly greater than that in systemic lupus erythematosus and in normal controls (P< 0.01 ).Conclusion:These results indicated that the autoantibody to HSP71 was main autoantibodies to HSPS in ISNHL and its presence might serve as a marker for an autoimmune etiology of these patients′ hearing loss.
