【摘要】 目的：探讨白细胞介素１３（ＩＬ－１３）对体外培养的大气系膜细胞的增殖及其产生白细胞介素１β（ＩＬ－１β）的影响。方法：用四甲基偶氮唑（ＭＴＴ）法测定系膜细胞增殖，用酶联免疫吸附（ＥＬＩＳＡ）法测定培养系膜细胞的上清液 ＩＬ－１β蛋白水平。结果：ＩＬ－１３在 １．０、１０、１００ ｎｇ／ｍｌ浓度范围呈剂量依赖性地抑制系膜细胞的增殖；含 ５％ＦＣＳ的ＲＰＭＩ１６４０培养状态下的系膜细胞几乎检测不出ＩＬ－１β分泌，加入ＬＰＳ可诱导系膜细胞分泌ＩＬ－１β，ＩＬ－１３在抑制系膜细胞的增殖的相应浓度范围可显著地抑制 ＬＰＳ诱导的系膜细胞 ＩＬ－ １β分泌。 ＩＬ－ １３在 １０ ｎｇ／ｍｌ浓度时即可几乎完全抑制 ＬＰＳ诱导的系膜细胞分泌 ＩＬ－１β。结论：ＩＬ－１３对体外培养的系膜细胞增殖具有抑制作用，ＩＬ－１３可能对肾小球系膜细胞炎症具有拮抗作用。更多还原

【Abstract】 Objective:To explore the effects of interleukin - 13 (IL- 13) on cell proliferation and interleukin - 1 β (IL - 1 β) secretion in mesangial cell. Method: Cell proliferation was tested by MTT method. The protein synthesis and secretion of IL - 1β in mesangial cells was measured by ELISA. Results: IL - 13, The proliferation of mesangial cells could be inhibited in the range of IL - 13 concentration from lug/ml to 100 ng/ml with or without inducing yb LPS. The protein release of IL - 13 also be inhibited obviously. IL - 13 in 10 ng/ml could inhibit almost completety IL -1β secretion induced by LPS. Conclusion: The results showed that IL - 13 could inhibit the proliferation of mesangial cells and IL -1ｗβ secretion induced by LPS in mesangial cells. It was suggested that IL - 13 may be an inhibitory cytokine in regulation of inflammatory reaction in glomerular mesangial cells.更多还原
