抗肠出血性大肠杆菌O157∶H7鸡卵黄抗体的初步研究
Preliminary Study on Chicken Egg Yolk Antibody against EHEC O157∶H7
【摘要】 [目的 ]制备高效价特异性鸡卵黄免疫球蛋白 (Yolk Immunoglobulin,Ig Y)抗体 ,探索肠出血性大肠杆菌(EHEC O15 7∶ H7)防治的新方法。 [方法 ]用 EHEC O15 7∶ H7菌株制备抗原 ,免疫育龄 L eghorn鸡 ,获取大量含高效价Ig Y的卵黄 ,经水稀释法脱脂、盐析、初步纯化浓缩 ,用 EL ISA检定其免疫学活性 ,并测定提取物蛋白含量。 [结果 ]获得EL ISA效价达 1× 10 - 6 的抗 O15 7∶ H7Ig Y抗体 ,其蛋白含量为 141.8mg/ ml。 [结论 ]用水稀释法制备 Ig Y抗体简便、经济、高效。 EHEC O15 7∶ H7菌株抗原可有效地导致种鸡免疫反应 ,用其卵黄制备的 Ig Y抗体产量多、效价高、特异性强 ,可进一步用于抗 EHEC O15 7∶ H7感染的保护性研究。
【Abstract】 Objective] To produce the specific and efficient chicken egg yolk immunoglobulin (IgY) in order to develop a new way for the prevention or immunotherapy against infectious disease of EHEC O157∶H7 [Methods] The antigen of EHEC O157∶H7 strain was used to immunize the hens and produce a large amount of high titer IgY which was separated from the granules and lipids in the egg yolk by employing water dilution method The immuno activity of IgY was detected by ELISA [Results] The protein content of purified IgY was 141 8 mg/ml The ELISA titer in the purified IgY was higher than 1×10 -6 [Conclusion] The egg yolk of immunized chicken is a rich and inexpensive source of specific antibody EHEC O157∶H7 strain antigen was effective to induce an immune response The IgY,which can be separated by water dilution method which had no adverse effect on the immunoactivity of IgY, would be used for further prevention study against EHEC O157∶H7 infection
