地高辛标记探针原位杂交法检测人脑胶质瘤组织猴病毒40的表达
Expression of simian virus 40 DNA in human brain glioma tissues
【摘要】 目的 :检测猴病毒 40 (SV40 )DNA在人脑胶质瘤中的定位表达 ,探讨其在脑胶质瘤发生发展中的生物学意义。方法 :采用地高辛标记探针原位杂交技术 ,检测 2 5 6例人脑胶质瘤和 11例正常脑组织标本。结果 :SV40DNA定位于胶质瘤细胞核 ,阳性细胞呈弥漫性或局灶性分布 ;SV40DNA阳性率 40 .6 % (10 4/2 5 6 ) ,正常脑组织均未检测出SV40DNA ;SV40DNA阳性率与病理分级无关 (P >0 .0 5 )。结论 :SV40感染与人脑胶质瘤病因学密切相关 ;地高辛标记探针原位杂交技术 ,是探讨SV40病毒核酸在组织中定位、分布和感染机制的一种新的特异性方法。
【Abstract】 Aim: To detect the location of simian virus 40 (SV40) DNA and its probable biological function in oncogenesis of human brain gliomas.Methods:A total of 256 cases of human brain glioma tissues and 11 cases of normal brain tissues were studied for SV40 DNA by digoxin labeled probe in situ hybridization.Results:Hybridization signals were detected within the nuclei of glioma cells and the positive cells distributed diffusely or focally.SV40 DNA positivity rate was 40.6% in brain glioma tissues,whereas none of the normal tissues had positive results.The correlation brain between SV40 DNA positivity rate and pathological degree of tumor was not apparent(P>0.05).Conclusion:SV40 infection is closely related to etiology of glioma.The technique of digoxigenin labeled probe in situ hybridization is a new specific method in studying the location,distribution and infection mechanisms of SV40 DNA.
