黑曲霉SL2-111复合酶固体发酵工艺研究
Solid fermentation of Aspergillus niger SL2-111 producing multi-enzymes
【摘要】 以酸性蛋白酶酶活为响应值 ,采用单因素搜索和正交试验对黑曲霉 (Aspergillusniger)SL2 111固体发酵工艺进行优化 ,结果表明最适培养基的组成为 :新鲜麸皮 8.2 5 g、米糠 4 .5g、豆饼粉 1.5 g、(NH4 ) 2 SO4 0 .3g、K2 HPO4 0 .0 6 g、CaCl2 0 .0 75 g、水 8.6mL ,pH5 .5 ,变温培养 ,前 30h2 8℃、后 30h为 2 3℃ ,培养时间为 6 0h。采用最适培养基和优化工艺 ,在 2 5 0mL三角瓶中进行验证实验 ,酸性蛋白酶酶活可达 12 5 86U /g ,果胶酶和纤维素酶分别为 16 4 90U/g、982 2U/g。
【Abstract】 The solid fermentation condition of Aspergillus niger SL2-111 was studied by using single-factor research and orthogonal test based on the response value of acid protease. The results showed that the optimal medium contained 8.25 g fresh bran, 4.5 g rice bran, 1.5 g cake bean, 0.3 g (NH4)2SO4, 0.06 g K2HPO4, 0.075 g CaCl2, 8.6 mL water. The initial pH was at 5.5, and culture temperature was fermented at 28℃ for 30 h and then at 23℃ for later 30 h. The enzyme activities of acid protease, pectinase and cellulase were 12586 U/g, 16490 U/g and 9822 U/g, respectively, when the strain was grown on the optimal medium under the above culture conditions in 250 mL Erlenmeyer flask.
