近似等位基因系小麦盐胁迫下叶绿体超微结构的比较研究

【摘要】 两个近似等位基因系 (RH870 6 4 9和H870 6 34 )在含有 1%NaCl的Hoagland溶液中分别培养36h、4 8h、60h ,再返回到无盐的Hoagland培养液中培养 36h ,用透射电镜对两品系在不同处理条件下叶绿体的超微结构进行了观察。结果表明 :随着盐胁迫程度的加剧 ,叶绿体受伤害程度加重 ,耐盐性较差的H870 6 34的叶绿体对盐胁迫更为敏感 ;盐胁迫解除后 ,耐盐性较强的RH870 6 4 9叶绿体的外部形态和内部结构都得到一定程度的恢复 ,耐盐性较差的品系则恢复不好或难以恢复。对两品系类囊体膜的损伤情况进行统计分析 ,发现RH870 6 4 9的类囊体膜损伤主要表现为微溶 ,H870 6 34主要表现为重溶 ,从细胞水平上为RH870 6 4 9的耐盐性优于H870 6 34提供了证据 ,为进一步研究耐盐机理提供了好材料。

【Abstract】 Two near isogenic lines (RH8706 49 and H8706 34)were cultured in Hoagland solution containing 1% NaCl for 36h,48h,60h respectively,and then restored to Hoagland solution without salt for 36h. The chloroplast ultrastructures of the two lines were observed under TEM with different treatments,the results showed that with the intensifying of salt stress,the chloroplasts were injured to a larger degree. The chloroplasts of H8706 34 with less salt tolerance were more sensitive to salt stress;after the removal of salt stress,the chloroplasts of the more salt tolerant can restore both in their morphology and internal structure,while they could not resume or difficult to restore in RH8706 34. Statistical analysis of the thylakoid membrane injury was also conducted in these two lines,the results showed that most of the thylakoids injured in RH8706 49 melt lightly,while in H8706 34,they melt seriously,which could provide evidence for the superiority of RH8706 49 to H8706 34 in salt tolerance on cell level and also provide good materials for further study on salt tolerance mechanism.
