非水相中脂肪酶转化失水山梨醇单油酸酯反应的研究
Lipase-Catalyzed Synthesis of Sorbitan Esters in Non-aqueous Phase
【摘要】 失水山梨醇单油酸酯可通过脂肪酶CAL在无溶剂体系中转化合成。对反应体系进行条件优化后 ,进行了10批酶法合成失水山梨醇单油酸酯 ,结果表明该酶具有较高的催化活性和稳定性 ,每批转化率均达 92 %以上 ,10批反应结束 ,酶活性仍有约 70 % ,半衰期为 92 1 4h。但单酯含量逐批略有下降 ,从最初 85 3 %降至约 5 6 6%。与化学合成的产品比较 ,各项指标基本接近 ,但酶法合成的产品前 5批单酯含量较高 ,均可达 70 %以上
【Abstract】 Esterification of sorbitan and oleic acid can be carried out by lipase CAL in nonaqueous phase under mild conditions.After the reaction conditions was optimized,the lipase catalyzed 10 batches of synthesis of sorbitan monooleate,both the catalytic activity and stability of the lipase CAL maintain a high lever.The conversion in every batch is more than 92 %.After 10 batches reaction,the remained activity of lipase CAL is about 70 %,and its half life is 921 4 h.However,the preparative yeld for sorbitan monoesters decrased gradually with batches,from 85 3 % at first to 56 6 %.A comparison was made for the products from different synthesis.The enzymaticly prepared sorbitan monooleate were similar to the commercial products in acid number,hydroxyl number,saponification number and color.However the content of sorbitan monoester in the first five batches enzymatic products was higher,up to about 70 % and more.
