东北虎中SOX基因的检测
THE DETECTION OF SOX GENES IN PANTHERA TIGRIS ALTAICA
【摘要】 应用特异于HMG bOX区域的兼并引物 ,扩增了东北虎的SOX基因。在扩增产物中发现五条大小分别为 2 2 0 ,2 70 ,3 5 0 ,4 3 0和 5 60bp的扩增带。经过与地高辛标记的人SRY基因进行Southern杂交表明这五条扩增带均呈现阳性 ,说明它们均为东北虎的SOX基因片段 ,这些基因保守区的长度在基因组DNA水平上存在着明显的差异。
【Abstract】 The SOX genes of Panthera tigris altaica Temminck were detected by a pair of degenerate primers. The degenerate primer were special to the SOX genes family within the HMG-box. With the genomic DNA as template, five bands were observed with the length of 220,270,350,430 and 560 bp. With the Dig-labeled human SRY gene as probe, Southern blotting results indicated that all the bands are specially positive. So the conserved box length of different SOX genes are different from each other in the genomic DNA. The reason of the length divergence in PCR products was discussed.
