靛酚蓝光度法测定蛋白质

【摘要】 提出一种测定蛋白质的新方法 ,样品与硫酸共热 ,蛋白质分解 ,其中的氮转为氨 ,氨由靛酚蓝光度法测定 .本法中有色化合物的最大吸收波长 62 0nm ,表观摩尔吸光系数ε62 0 =1.8× 10 4 mol- 1·L·cm- 1,线性范围 :0~ 0 .6μg/ml氮 ,检出限为 5.8× 10 - 6g/L氮

【Abstract】 A new method for determination of proteins was described. The sample was heated with sulphuric acid. Proteins in sample were decomposed and their nitrogen was changed into ammonia, that was determinated by indigo phenol blue spectrophotometry. The maximum absorption wavelength of colored complex was found to be at 620nm with apparent molar absortion cofeeicient of 1.8×10 4mol -1 ·L·cm -1 and the liner range was 0~0.6μgN/mL and sensitivity limit was 5.8×10 -6 gN/L.
