吸烟诱发人血淋巴细胞DNA氧化损伤的研究
DNA OXIDANT DAMAGE INDUCED BY TOBACCO SMOKE IN HUMAN BLOOD LYMPHOCYTES
【摘要】 目的 :研究吸烟对人血淋巴细胞DNA氧化损伤的作用。方法 :采用高效液相色谱系统结合电化学测定分析法对吸烟和不吸烟的男性健康人外周血淋巴细胞DNA中氧化损伤的生物标志物— 8 羟基 2 脱氧鸟苷 (8 OHdG)的含量进行分析。 结果 :①吸烟者比不吸烟者淋巴细胞DNA中的 8 OHdG显著增高 (P <0 .0 5 ) ,且 8 OHdG的含量与每天吸烟支数呈正相关 ;②吸烟对DNA氧化损伤的效应个体差异较大 ,本测定方法可用于对吸烟高敏感人群的筛选。 结论 :吸烟可引起人血淋巴细胞DNA氧化损伤
【Abstract】 Purpose: To investigate the oxidative damage of DNA in human blood lymphocytes. Methods: Levels of 8 hydroxy 2 deoxyguanosine (8 OHdG) in human blood lymphocytes from healthy male smokers and non smokers were determined by high performance liquid chromatography system with an electrochemical detector. Results: (1)the mean level of 8 OHdG was significantly higher in smoker group than non smoker group, and a positive correlation between content of 8 OHdG and number of cigarette smoking per day was found; (2) A large interindividual variation in 8 OHdG levels was observed in smokers. Conclusion: Tobacco smoke can caused oxidant damage to DNA in human blood lymphocytes.
