水溶性苦瓜多糖的提取与测定

【摘要】 苦瓜是一种传统药食同源瓜果 ,通过沸水提取、醇析、脱蛋白和透析 ,从鲜苦瓜中分离出多糖粗品 ,粗品经sephadexG 1 0 0柱层析纯化 ,硫酸 酚法鉴定出多糖部位 ,收集单一峰组分得一苦瓜多糖样品 ,分子量约 80 0 0 ,并利用薄层色谱和气相色谱对该多糖中的成分单糖进行了定性和定量分析 ,结果表明 ,苦瓜水溶性多糖是由鼠李糖、阿拉伯糖、甘露糖、葡萄糖和半乳糖组成的杂多糖 ,其百分组成分别为 2 4 0 3% ,1 6 47% ,6 7% ,2 4 84% ,2 7 94% ,红外光谱显示该多糖有 881cm 1 吸收峰 ,提示含有 β D 吡喃糖苷键 ,可能是一活性多糖

【Abstract】 Momordica charantia is a traditonal herbal medicine and vegetable.A water soluble polysaccharide is isolated and purified from fresh momordica charantia L by extraction with boiling water,precipitation with alcohol,removal of protein,and dialysis against water,then,further purification by chromatography on Sephadex G 100 column.The molecular weight of the water soluble polysaccharide is about 8 000.At the same time,IR spectrum of the white gourd polysaccharide has an absorption band at 881 cm -1 ,due to β glycoside linkage,this result shows the polysaccharide may be active component.The component sugars of water soluble polysaccharide are identified and quantified by thin layer chromatography(TLC)and capillary gas chromatography (CGC),the result shows that water soluble polysaccharide is made from rhamnose,arabinose,mannose,glucose,galactose with the contents of 24.03%, 16.47%, 6 70%,24.84% and 27.94% respectively.
