【摘要】 目的:探讨扶正化瘀方对大鼠急性CCl₄损伤肝Kupffer细胞功能的影响。方法:体外分离培养大鼠息性CCl₄损伤肝Kupffer细胞,制备扶正化瘀方药物血清对其进行干预,观察药物对其PDGF和TGFβ活性以及对Kupffer细胞线拉体呼吸功能的影响。结果:药物组PDGF和TGFβ活性均显著降低（P<0.05）;药物组线粒体呼吸功能明显降低（P<0.05）。结论:扶正化疼方对大鼠急性CCl₄损伤肝Kupffer细胞活化有抑制作用,可能抑制肝星状细胞旁分泌激活,是该方抗肝纤维化的作用机理之一。更多还原

【Abstract】 Aim: To investigate the effects of FZHYF on hepatic Kupffer cells in vitro. Method: Drug sera were collected from normal rats which administrated with FZHYF or normal salt as control and sera pharmacology was employed. Kupffer cells were isolated from the rat liver acutely injured by CCl4, these Kupffer cells were then treated with drug sera and the Kupffer cell conditioned culture medium (KCM) was collected. The KCM was tested by their effects on the ’proliferation of Mv-1-LU cell line and L929 cell line, which imply the activity of TGF β and PDGF respectively. The drug sera was also incubated with Kupffer cells to assy its reduction of MTT, which respect the activiation of Kupffer cell. Result: At 20% concentration, KCM from FZHYF drug sera treated Kupffer cells had an obviously lower activity of TGF β (23. 8%±9. 72%) than ontrol (42. 5%±7. 82%) (P<0. 05); at 20% and 30% , drug sera also showed a significant inhibition on PDGF activity of KCM (26. 63±12. 47 and 23. 19±16. 27 respectively) when compared with control (54. 08±11.06 and 57. 07±14. 94 respectively) (P<0. 05); and the MTT reduction of Kupffer cell was suppressed by the drug sera (1. 036±0. 074 ) significantly (P<0.05), while the control was 1. 224 ±0. 096. Conclution: FZHYF can inhibite the activity of TGF β and PDGF derived from the Kupffer cell of acute injured rat liver induced by CCl4; and can also suppress the Kupffer cell reduction of MTT. These results indicate that FZHYF may has an action against Kupffer cell activiation.更多还原
