【摘要】 通过细胞体外培养观察急性CCl4 肝损伤的大鼠肝库普弗细胞对正常肝星状细胞的激活作用。方法　以 5 0 %CCl4 橄榄油一次灌胃复制大鼠急性肝损伤模型 ,分离模型大鼠肝库普弗细胞并原代培养 ,收集条件培养液 ,以其温育正常大鼠分离的原代培养肝星状细胞 ,二苯基四氮唑溴盐 (MTT)法测定细胞增殖 ,ELISA法测定Ⅰ型胶原分泌。结果　损伤肝库普弗细胞条件培养液可显著促进肝星状细胞的增殖及Ⅰ型胶原分泌 ,并具一定量效、时效关系 ;还可促进星状细胞表型向肌成纤维样细胞转化 ,而正常大鼠分离的肝库普弗细胞则无明显作用。结论　肝损伤时库普弗细胞对肝星状细胞的活化有多方面旁分泌激活作用更多还原

【Abstract】 Objective To investigate the activation of hepatic stellate cell(HSC) by Kupffer cell from CCl 4 induced da maged rat liver.Methods Rats were treated with CCl 4 to induce acute liver injury, the hepatic Kupffer cells were then isolated from the rats and primary cultured, the cultured medium was collected as the Kupffer cell conditioned culture medium(KCCM), and incubated with HSC from the normal rat, the proliferation of HSC was assayed by MTT colorimetric test, and collagen I secretion mesured by ELISA.Results KCCM from the injured liver promote the phenotype of HSC transformed to myofibroblast like cell(MFB), and markedly enahanced the proliferation and the collagen I secretion of HSC.Conclusion KCCM has multiple paracrine effects on the activation of cultured HSC.更多还原
