白细胞介素-2基因对丙型肝炎病毒E2基因免疫效果的调节作用

【摘要】 目的 研究白细胞介素 2 (IL 2 )基因对丙型肝炎病毒 (HCV)E2基因免疫小鼠后效果的调节作用。方法 构建Ⅱ型HCVE2基因的真核表达载体p3E2 ,免疫组化法检测其在哺乳动物细胞中的表达情况。用p3E2单独或连同IL 2真核表达质粒一起对BALB/c小鼠进行尾部皮下注射 ,酶联免疫吸附试验 (ELISA)法检测E2抗体的产生情况。结果 E2蛋白在哺乳动物细胞中得到瞬时表达。应用p3E2单独或连同IL 2真核表达质粒一起免疫小鼠均可刺激小鼠产生E2抗体 ;协同注射IL 2基因的小鼠产生的抗体滴度明显高于单独注射p3E2组的小鼠。结论 协同应用IL 2基因可明显加强小鼠对HCVE2基因的免疫反应 ,为HCV基因疫苗的可行性提供了一定的支持依据

【Abstract】 Objective To investigate the modulation effect of interleukin 2 (IL 2) gene on the immune response to hepatitis C virus (HCV) E2 gene when codelivered. Methods An expression vector of HCV E2 gene (p3E2) was constructed and its expression was detected in mammalian cells by transient expression. BALB/c mice were given injections of p3E2 only or together with IL 2 gene intraepidermally once or twice, and the induced humoral immune responses were evaluated. Results Immunohistochemistry showed that the constructed vector was successfully expressed in mammalian cells. Mice immunized with both p3E2 and IL 2 genes exhibited a significant increase in the production of anti HCV E2 antibody compared with the mice immunized only with p3E2. Conclusion Humoral immune responses of BALB/c mice to HCV E2 antigen can be enhanced when HCV E2 gene and IL 2 gene are coinjected.
