C-sis反义寡核苷酸局部干预抑制主动脉损伤后内膜增生

Local delivery of C-sis proto-oncogene antisense oligodeoxynucleotides inhibits intimal hyperplasia in rabbit injured aorta

【摘要】 目的 研究主动脉球囊损伤后 ,C sis原癌基因反义寡核苷酸 (AODN)局部干预对血管内膜增生的影响。方法  3 6只新西兰白兔随机分为C sis原癌基因AODN组、正义寡核苷酸 (SODN)组及生理盐水组 ,每组 12只。球囊损伤胸主动脉后 ,1mgAODN、SODN用于损伤局部干预 ,高胆固醇饲养 ,用RT PCR技术及组织学方法结合图像分析 ,观察AODN对损伤血管C sis表达及内膜增生的影响。结果 AODN明显抑制损伤血管C sis表达 ,SODN无抑制作用 ;AODN组、SODN组及生理盐水组 ,内膜增生厚度分别为 (8 5 4± 1 5 9) μm、(2 4 5 9± 2 14 ) μm、(2 5 15± 2 3 1) μm ,AODN组平均抑制(65 7± 9 6) %的内膜增生。结论 C sis原癌基因AODN局部干预明显抑制球囊损伤血管内膜增生 ,其抑制作用与抑制损伤血管C sis原癌基因表达密切关联。

【Abstract】 Objective To study the inhibitory effect of locally delivered C sis proto oncogene antisense oligodeoxynucleotides (AODN) via balloon catheter on intimal injury following balloon angioplasty in rabbit aorta. Methods Thirty six rabbits were divided into AODN and sense oligodeoxynucleotides (SODN) treated groups and control group randomly ( n =12, each). Each of the rabbits was injured with channeled balloon catheter 1 mg of C sis proto oncogene AODN or SODN was locally delivered on injured aorta with inflated balloon at 0.5×10 3 kPa for 8 minutes. The rabbits were fed with 2.0% cholesterol diet for 3 days or 28 days. Expression of C sis proto oncogene was determined by using of RT PCR. Morphological analysis of intimal hyperplasia was performed with histologic staining and image analyzer. Result AODN inhibited C sis proto onconge expression in injured aorta obviously. SODN showed no effect on c sis mRNA. Thickness of intimal hyperplasia following locally delivered AODN, SODN and 0.9% NaCI solution on injured aorta was 8.54±1.59, 24.59±2.14 and 25.15±2.31 μm, respectively. Inhibitory rate of intimal hyperplasia with locally delivered AODN was (65.7±9.6)%. Conclusion Local delivery of C sis proto oncongene AODN following balloon angioplasty on injured aorta inhibits intimal hyperplasia, and the inhibitory effect was associated with suppression of c sis proto oncongene expression on the injured aorta.
