TGF-β1及其受体在两种人肾癌细胞系中的表达

【摘要】 目的 探讨转化生长因子 β1(TGF β1)及其受体在人肾癌细胞系和正常肾细胞中的作用。 方法 应用免疫细胞化学 (LSAB)和RT PCR方法 ,检测二种人肾癌细胞系 (GRC 1和RCC 949)和一种原代培养肾纤维母细胞 (PRF)TGF β1及其Ⅰ和Ⅱ型受体的表达情况。 结果 三种细胞均表达TGF β1和I型受体 ,其中RCC 949表达Ⅱ型受体量较原代培养肾纤维母细胞减少 ,而GRC 1则不表达。 结论 由于人肾癌细胞系表达Ⅱ型受体量减少或丢失 ,因而对TGF β1抑制作用减弱 ,这可能是肾癌细胞系呈侵袭性生长的部分原因。

【Abstract】 Objective To study the effects of transforming growth factor β 1 (TGF β 1)in renal carcinoma cells and in normal culture renal cells. Methods Expressions of TGF β 1,TβR 1 and TβR Ⅱ in 2 kinds of human renal carcinoma cell lines and in primary cultured renal fibroblasts were studied by means of immunocytochemistry and PCR technique. Results All the 3 groups have expressions of TGF β 1 and TβR I. Whereas TβR Ⅱ has been absent in one of the human renal carcinoma cell line and lower in the other. Conclusions The results demonstrated that human renal carcinoma cell lines are insensitive to the inhibitory effect of TGF β 1 due to reduction or loss of TβR Ⅱ. This phenomenum might be responsible for the aggression of the renal carcinoma cells.
