脑肿瘤患者脑脊液中基质金属蛋白酶活性表达的研究

【摘要】 目的 :研究肿瘤组织中的基质金属蛋白酶 ( matrix m etalloproteinase,MMPs) ,尤其是明胶酶 A( MMP-2 )和 B( MMP- 9)的活性与原发及转移性脑肿瘤恶性程度和浸润能力的相关性。方法 :选取 6 4例脑肿瘤病人及 14例对照组脑脊液 ,利用明胶 Zymography法检测 MMP- 2和 MMP- 9的表达及活性差异。结果 :原发性和转移性脑肿瘤中明胶酶 A和 B活性表达率较对照组均有显著差异 PA<0 .0 1,PB<0 .0 5。间变性脑膜瘤组与良性脑膜瘤组之间明胶酶 A活性表达率有显著差异 ( P <0 .0 5 )。结论 :对脑肿瘤病人的脑脊液中明胶酶的活性分析可在一定程度上反映脑肿瘤的恶性程度。

【Abstract】 Objective:The purpose was designed to investigate the relationship between specific matrix metalloproteinase (MMPs), particularly gelatinase A and B in human cerebrospinal fluid with brain tumor with the grade and aggressiveness of primary and metastatic brain tumors.Methods:Cerebrospinal fluid(CSF) from 64 patients and 14 controls were analyzed for the presence of gelatinolytic activity by zymography. Results:CSF from all patients with malignant brain tumors or brain metastases contained MMP 2 and MMP 9 whereas control CSF contained none of them. Conclusion:The activated forms of gelatinase A and B can be detected in the CSF of patients with primary and metastatic brain tumors. The distribution of gelatinases activity in CSF might represent the malignant degree and invasive ability of brain tumors.
