【摘要】 本试验用绵羊红细胞免疫鸡 ,免疫后 5天采血 ,制备富集IgM血清。应用硫酸铵粗提、Sepharose₄B分子筛层析方法 ,从富集IgM的鸡血清一次性分离提取高纯度的IgM及纯度较高的IgG ,并经DEAE₅ 2纤维素离子交换层析进一步纯化 ,获得电泳纯的IgG。经鉴定该方法制备的IgG ,IgM完全可以满足免疫学实验要求 ,从一份血清样品中同时纯化IgM、IgG两种免疫球蛋白 ,既省时又节省原材料更多还原

【Abstract】 IgM and IgG were prepared from the IgM enriched chicken serum by precipitation with 50%,33%,33% staturated anmonium sulphate,gel filtration on Sepharose₄B.IgG preparation was chromatographed a second time on a DEAE_cellulose exchange.Analysis by SDS_PAGE and Western_blot confirmed successful purification of chicken IgM and IgG.The method is inexpensive and rapid.更多还原
