【摘要】 本试验首先用PCR方法扩增出ILTVgB基因 ,将其克隆到质粒pUC1 1 9中 ,并进行了序列分析。将克隆的gB基因再亚克隆到禽痘病毒转移载体质粒中 ,然后 ,通过质脂体方法转染由禽痘病毒感染的鸡胚成纤维细胞 (CEF)中 ,筛选蓝色的重组病毒 ,并对其进行了 6轮蚀斑纯化。该重组病毒通过PCR方法鉴定证明其基因组中含有完整的鸡传染性喉气管炎gB基因 ,Westernblot实验证明了该重组鸡痘病毒表达了鸡传染性喉气管炎gB糖蛋白更多还原

【Abstract】 A 2.7Kb DNA fragment encoding the glycoprotein B(gB) of infectious laryngotracheitis virus wangguang strain,a virulent isolate of China,was obtained by PCR and sequence was determined.The 2619 base_pair sequence encodes a protein of 873 amino acids.The gB gene of ILTV and LacZ gene of Ecoli,under the control of the fowlpox Lp2Ep2 promoter and the vaccinia P11 promoter respectively, were inserted into a nonessential region of fowpox virus.The recombinant fowlpox virus (rFPV) were identified by blue_plaque cloning and PCR method.A glycosylated B protein expressed by the rFPV were recognized by anti_ILTV chicken serum on the western blot.Experiments are in progress to test the protective ability of the rFPV against fowlpox disease and infectious laryngotracheitis.更多还原
