新城疫病毒四平株HN蛋白基因的克隆与序列分析
Cloning and Sequence Analysis of HN Protein Gene of Newcastle Disease Virus Strain Siping
【摘要】 新城疫病毒 (NDV)四平株在鸡胚增殖后纯化 ,提取RNA ,然后利用特异性引物 ,经RTPCR一次性扩增出了NDV四平株的全长HN基因。将该HN基因插入pKS()后测序。序列分析表明 ,该HN基因核苷酸长度为 1 742bp ,编码 5 71个氨基酸 ,序列中有 5个糖基化位点 ,1 2个半胱氨酸残基。同源性分析表明 ,四平株HN基因与目前国外发表的其他NDVHN基因相比 ,核苷酸序列同源性在 83.4%89.2 %之间 ,推导的氨基酸序列同源性在 87.6 %91 .1 %之间。
【Abstract】 The NDV strain Siping was grown in chicken embryos. The virus was purified and the genomic RNA was extracted. Using a pair of special primers,whole sequence of HN gene of NDV strain Siping was amplified by RT_PCR and sequenced after being directly inserted in plasmid KS(_).The sequence analysis showed that the nucleotide sequence of this HN gene was 1742 bp long encoding a protein of 571 amino acids. In the amino acid sequence,there are 5 glycosylation sites and 12 cysteine residues.The homology of the nucleotide is 83.4%~89.2% and that of the deduced amino acid sequence is 87.6%~91.1%.
