线粒体DNA与细胞癌变关系研究进展
Research advances in relationship between mitochondrial DNA and carcinogenesis of mammalian cells
【摘要】 人类细胞线粒体DNA (mitochondrialDNA ,mtDNA)是一具有 16 5 6 9个碱基对的双链闭环分子 ,它包含 2 2个tRNA和 2个rRNA基因 ,并编码 13个有关细胞线粒体氧化磷酸化的蛋白多肽。与核DNA相比 ,mtDNA分子更容易受致癌物的损伤。提示细胞质遗传与核遗传系统共同参与了细胞癌变过程。本综述重点讨论mtDNA损伤与突变在细胞癌变中的地位和可能的作用机制
【Abstract】 Human mitochondrial DNA (mtDNA) is a 16?569 base pair double stranded,closed circular molecule,which codes for two ribosomal RNAs,22 transfer RNAs and 13 proteins. All of the mitochondrial genes encode subunits of the oxidative phosphorylation (OXPHOS) enzymes that are responsible for the energy generating pathway. mtDNA is thought to be more susceptible to damage by mutagens than nuclear DNA. This implies that both nuclear and cytoplasmic elements are responsible for the steps of carcinogenesis.So,in this article,we reviewed the relationship between mtDNA and carcinogenesis of mammalian cells. The possible mechanisms were also discussed.
