用SCGE法检测燃煤污染型砷中毒患者血细胞DNA的损伤作用
Detection of DNA damage in blood cells of the patients of coal burnt arsenism by SCGE
【摘要】 目的 探讨砷中毒的致癌机理及寻找灵敏、实用的 DNA损伤监测指标。方法 应用单细胞凝胶电泳法 (SCGE)对贵州省兴仁县燃煤型砷中毒重病区 175名砷接触者的血细胞 DNA损伤进行了检测 ,并分析了相关因素的影响。结果 砷能引起人体血细胞 DNA单链断裂 ,其损伤发生早于临床表现 ,并与尿砷、发砷呈正相关关系 :与皮肤损害进展一致。吸烟和氟的影响可加重砷对 DNA的损伤作用。结论 SCGE具有灵敏、快速、简便等优点 ,可作为一种早期、实用的方法应用于砷接触人群的 DNA损伤监测
【Abstract】 Objective In order to research arseniasis’s mechanism on causing poisoning,especially resulting in skin carcinoma and search sensitive,practical monitoring index of DNA damage.Methods Using single cell gel electrophoresis assay (SCGE),the DNA damage in blood cells of 175 arsenic contacts was detected in Xingren County,Guizhou Province,where there was serious arsenism caused by burning coal,and the effects of related factors were analyzed too.Results Arsenic could induce DNA single strand breaks in human blood cells,which might happen before the occurrence of clinical manifestations.The DNA damage caused by arsenic had a positive correlation with the amount of arsenic in urine or hair,and coincided with the development of the pathological changes of skin.Cigarette smoking and fluorine might aggravate the arsenic induced DNA damage.Conclusions It is suggested the SCGE can be used as a sensitive、rapid、handy、practical and early stage monitoring method to detect DNA damage in the population exposed to arsenic.
