【摘要】 目的 研究胚胎肝发育相关基因 1(EDAG 1)在红系肿瘤分化中的表达及意义。方法 用细胞生物学、RT PCR及Northern杂交法。结果 含 4 0× 10 -5mol·L-1氯高铁血红素 (Hemin)的RPMI 16 40培养K5 6 2细胞 5天 ,可使70 %~ 80 %细胞呈现正常红系特征 ;EDAG 1、c fos表达降低。结论 Hemin可诱导红白血病细胞分化 ;EDAG 1、c fos表达下调可能与其有关。

【Abstract】 Objective To study the expression and role of EDAG-1 in erythroleukemia differentiation induced by Hemin. Methods Techniques of cellular biology were used to analyse K562 cells differentiation; Northern blotting and RT-PCR were used to study the expression of EDAG-1 and c-fos gene. Results 70%~80% K562 cells were induced to differentiate to erythrocytes by 40×10 -5 mol·L -1 Hemin; There was slower proliferation in these cells; the expression of the EDAG-1 and c-fos decreased in differentiated K562 cells. Conclusion K562 cells can be induced to differentiate by Hemin. EDAG-1 and c-fos may be involved in the process.
