帕金森病基因治疗的研究进展

【摘要】 基因治疗帕金森病(Parkinson’sdisease,PD)的研究是神经科学领域的热点之一。靶基因的导入可以增加多巴胺的合成 ,降低多巴胺神经元的死亡。靶基因编码多巴胺合成途径中重要的酶如酪氨酸羟化酶(TyrosineHydroxylase,TH)和芳香族氨基酸脱羧酶(AromaticL -AminoacidDecarboxylase,AADC)以及神经元保护分子如神经营养因子和抗凋亡蛋白。基因的导入主要有2种方式 :直接转移(InVivo)和离体转移(ExVivo)。本文综述了不同的基因转染系统。虽然目前PD的基因治疗还停留在实验室阶段 ,但现有的结果是令人鼓舞的。

【Abstract】 Gene therapy is being explored as an effective strategy to restore and protect neuronal function in the substantia nigra.Candidate genes include those that either can augment dopamine biosynthesis such as tyrosine hydroxylase(TH)and aromatic L_amino acid decarboxylase(AADC)or attenuate loss of dopaminergic neurons such as neurotropic factors and anti_apoptosis proteins,Two primary approaches for gene transfer have been explored :in vivo and ex vivo.Various gene transfer systems have been postulated .Although all of them are still at the stage of laboratory experiment,the attained results appear quite promising.
