【摘要】 目的 对软下疳病原体杜克雷嗜血杆菌的运输、分离及保存培养基进行评价和筛选,建立中国软下疳的实验室诊断方法,为开展软下疳的流行病学调查和监测提供必要的手段。方法 通过接种培养杜克雷嗜血杆菌标准菌株对其菌落数量及大小进行评价,筛选出最适目的培养基。结果 杜克雷嗜血杆菌标准菌株:(1)在增养淋球菌培养基和碳琼脂培养基中经过3次传代均能保持良好生长;(2)在改良的Stuart培养基中,置4℃ 9天,仍能保持其活性便于运输;(3)在二甲基亚砜和甘油保存培养基中-70℃保存180天,活性大于脱脂牛奶培养基。结论 研究表明增养淋球菌、碳琼脂两种培养基,改良的Stuart培养基及二甲基亚砜和甘油培养基分别为杜克雷嗜血杆菌的最佳分离、运送及低温保存培养基。更多还原

【Abstract】 To establish the diagnostic procedure of the nationally chancroid laboratory to provide the prerequisite for carrying out the epidemiological research and surveillance to it, the isolating, transporting and storing media of its strain, Haemophilus ducreyi (Hd) , were evaluated and screened. Methods Three kinds of isolating media and two transporting and three storing ones were evaluated and selected as the most appropriate media by being inoculated and cultured the standard strain of Hd provided by Laboratory Center of CDC, Canada, and being measured the quantity and size of its colonies. Results (1) Hd could keep growing well in the enriched carbonic agar medium after three times of inoculation; (2) In the modified Stuart medium for transporting Hd its viability had remained for 9 days at 4** , and the dimethyl sulfoxide and glycerol storage media had preserved it for more than 180 days at - 70℃ with a higher viability of it than that in the de - fatted milk medium. Conclusion The enriched carbonic agar medium, the modified Stuart medium, the dimethyl sulfoxide and glycerol storage media are the best media of Hd being isolated, transported and stored at low temperature, respectively.更多还原
