银染端粒重复扩增法的建立及初步应用
Establishment of Silver-Staining TRAP and Its Primary Application
【摘要】 目的 建立检测端粒酶活性的银染端粒重复扩增法。方法 以银染替代放射性核素分析结果 ,加入内标防止假阴性结果出现而建立银染端粒重复扩增技术 ,并运用此法检测 36例乳腺癌组织和 2 4例乳腺良性病变组织标本端粒酶活性 ,同时与溴乙锭染色的端粒重复扩增法进行比较。结果 36例乳腺癌组织中 33例阳性 (91.7% ) ,2 4例乳腺良性病变组织无 1例阳性。与溴乙锭染色端粒重复扩增技术相比 ,银染端粒重复扩增技术可见到更多更清晰的带形。结论 实验结果表明本法与放射性同位素显影的端粒重复扩增技术相比是 1种更为简便、安全、可靠的端粒酶活性检测方法。
【Abstract】 Objective To establish a silver-staining telomere repeat amplification protocol for detecting telomerase activity.Methods We established a silver-staining TRAP by replacement of radioisotope with silver- staining and introduction an internal standard.Telomerase activity of 36 breast carcinoma specimens and 24 benign breast lesions was analyzed with the method.This method was compared with the ethidium bromide-staining telomere repeat amplification protocol.Results Telomerase activity was detectable in 33 of 36(91.7%) breast cancer and in 0 of 24 benign breast lesions.The silver-staining telomere repeat amplification protocol could display more and clearer telomerase band compared with the ethidium-staining telomere repeat protocol.Conclusion The results suggested that the method is a simple,convenient,safe and reliable method to detect telomerase activity.
