MTS/PMS比色法测定血清黄嘌呤氧化酶
Colormetric Assay of Serum Xanthine Oxidase Using the Tetrazolium Salt MTS/PMS
【摘要】 建立了测定黄嘌呤氧化酶的MTS/PMS还原法。用黄嘌呤作为底物 ,酶促反应过程中生成的超氧离子将MTS还原成呈色的甲月替 ,生成的甲月替为水溶性 ,在 490~ 580nm间有一平坦的吸收峰 ,其ε50 0为 2 738× 1 0 4 。该方法具有良好的重复性 (XOD :2 38u/L ,CV4 31 % ;XOD30 8u/L ,CV2 81 % )和线性。参考值为 1 0 2 4± 0 82u/L。
【Abstract】 The method for determination of Xanthine Oxidase(XOD) by means of the reduction of MTS 〔3-(4 5 dimethylthiazol-2-yl)-5-(3-carboxy-methoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium,inner salt〕 was established with xanthine as the substrate.The MTS was reduced to an aqueous soluble formazan by superoxide produced in the enzymic reaction.The maximum absorbance of the formazan is at 490~580 nm,and its ε 500 is 2 738×10 4。This method has a good reproductivity(XOD2 38 U/L, CV 4 31%;XOD30 8 U/L, CV 2 81%)and wide linearity range.The normal reference range of the method is 1 024±0 82 U/L。
