花生大豆种子中SOD同工酶的研究

【摘要】 花生、大豆种子中的 SOD粗提液经聚丙烯酰胺凝胶电泳及 NBT活性染色 ,结果表明 ,花生种子有 6条SOD同工酶谱带 ,其中 1条为 Mn- SOD,其余为 Cu,Zn- SOD。大豆种子有 8条 SOD同工酶谱带 ,其中 3条为 Mn-SOD,其余为 Cu,Zn- SOD。不同 p H值对 SOD活性有一定的影响 ,p H =1.0时 ,Mn- SOD同工酶谱带消失 ;p H =12 .0时 ,全部 SOD同工酶谱带消失。变性剂 DTT、SDS和 β-巯基乙醇对 SOD同工酶有较大影响 ,SDS使 Mn- SOD谱带丢失 ,DTT、β-巯基乙醇使全部 SOD同工酶谱带消失

【Abstract】 SOD crude extracts from seeds of peanut and soybean were used for analysis by polypropylenegel electrophoresis and NBT active chromatid.It were shown that there were six bands in the isoenzymes zy- mogram of SOD for peanut seeds,in which one of them was Mn- SOD and others were Cu,Zn- SOD;there were eightbands for soybeam seeds,three ofwhich were Mn- SOD,the restwere Cu,Zn- SOD.The SOD ac- tivities was affected by p H value of crude extract.The Mn- SOD isoenzymes bands was lacked when p H was 1 .0 .All bands were disappered when p H was 1 2 .0 Denaturants DTT,SDS andβ- mercapto ethanol affected greatly SOD isoenzymes zymogram.SDScould lead to loss of Mn- SOD band,DTT andβ- mercapto ethanol set off loss of all SOD isoemzymes.
