纤维连接蛋白对大鼠肺成纤维细胞纤维连接蛋白和整合素α5β1表达的影响
Influence of Fibronectin on the Expression of Fibronectin and Integrin α 5β 1 in Rat Pulmonary Fiboblasts
【摘要】 目的 研究纤维连接蛋白 (fibronectin ,FN)及其受体整合素α5β1在肺纤维化中的作用。方法 应用Northern印迹杂交和免疫细胞化学技术。结果 经FN作用 2h ,肺成纤维细胞α5β1和FNmRNA的杂交信号均增强 ,6h达最高值 ;FN作用 6、1 2、2 4h ,免疫细胞化学染色显示整合素α5β1、FN蛋白的表达增强 ;经α5单克隆抗体预作用 ,FN作用组的肺成纤维细胞整合素α5β1和FN蛋白表达减弱 ,其抑制作用依次分别在 6h(α5β1)、1 2h(FN)最明显。结论 FN能直接刺激肺成纤维细胞合成FN或通过上调整合素α5β1的表达来促进自身合成FN。
【Abstract】 Purpose To study the influence of fibronectin (FN) on the expression of FN itself and its receptor,integrin α 5β 1 in pulmonary fibrosis. Methods Northern blot analysis and immunocytochemical stain were used on cultured rat pulmonary fibroblasts (PFBs) after FN treatment. Results The hybridization signals of α 5,β 1 and FN mRNA of PFbs enhanced at 2 h after FN treatment,and reached their peaks at 6 h.The expression of α 5β 1 and FN proteins increased after 6 h of FN treatment.Anti α 5 monoclonal antibody could partly inhibit the effect of FN on the expression of integrin α 5β 1and FN proteins in PFbs.The most distinctive inhibition was detected at 6 h(α 5β 1) and 12 h(FN) respectively. Conclusions FN could enhance the synthesis of FN of PFbs directly,or indirectly by the enhancement of the expression of integrin α 5β 1.
