狼毒大戟水提物对小鼠致突变作用的实验研究

【摘要】 目的 :探讨狼毒大戟水提物是否具有致突变作用。 方法 :实验用昆明种近交小鼠为实验对象 ,采用传统的毒理学实验方法 ,以微核出现率和精子畸变率为观察指标 ,在高 ( 360 g/kg)、中 ( 1 80 g/kg)、低( 30 g/kg)剂量下 ,进行了初步观察。 结果 :中、高剂量组与阴性对照组间有显著性差异 ( P<0 .0 1 ,P<0 .0 0 5) ,低剂量组与阴性对照组间无显著性差异 ( P>0 .1 0 )。结论 :提示狼毒大戟水提物在较高剂量下有致突变作用 ,但在低剂量下 ( 1 0倍于治疗剂量 )无明显毒性。

【Abstract】 Objective: To investigate the potential effect of Euphorbia Fisheriana Stead(EFS) on developing mutation.Methods: The present study was performed in inbred Kunming mice, using the traditional toxicological method based on observing ratios of micronucleus and sperm abnormity(RMS). The mice were randomly divided into 5 groups: negative control(Ⅰ) and positive control(Ⅱ), low dosage(Ⅲ), medium dosage(Ⅳ), and high dosage(Ⅴ), and the EFS extract 30g/kg, 180g/kg, 360g/kg were administered to groups Ⅲ, Ⅳ, Ⅴ respectively.Results: The results showed that high and medium dosages could significantly increase RMS as compared to negative controls(P<0.01,P<0.005),but similar result couldnt be observed in group Ⅲ(P>0.10).Conclusion: The reuslts indicate that EFS can cause mutation in medium and high dosage, whereas low dosage(10 times higher than that used in treatment) is obviously secure.
