RAPD标记在分析家鸡遗传变异中的应用
THE USE OF RANDOM AMPLIFIED POLYMORPHIC DNA(RAPD)FOR DETECTION OF POPULATION GENETIC VARIATION IN CHICKEN
【摘要】 作者选用 2 0个RAPD引物 ,分析了 8个家鸡品种 (系 )及两个杂交群体的遗传差异、群体内杂合度 ,并用计算出的遗传距离对 10个家鸡群体进行了UPGMA聚类分析 .试验结果表明 ,筛选出的 2 0个引物有 15个可以重复并具有多态性 ,共检测了鸡基因组 84个位点的遗传变异 ,平均每个引物产生的多态性条带平均为 4 .1条 .由RAPD试验得到的群体间遗传相似系数和遗传距离与实际情况接近 ,聚类分析表明 ,RAPD标记可以揭示群体间的亲缘关系及遗传变异 .
【Abstract】 The genetic variations and heterozygosities of 8 chicken breeds (or lines) and 2 hybrid chicken populations were analyzed by using 20 RAPD primers in this study,UPGMA cluster analysis based on the 10 chicken populations was performed applying the genetic distances. The results from this study indicated that 20 markers selected from 150 RAPD markers only 15 could produce reproducible and polymorphic results, a total of 84 loci’s variations in chicken genome were detected by the 15 markers, the average reproducible major bands produced by a RAPD primer were 5.6 including 4.1 polymorphic bands.The similarity coefficients and genetic distances between every two populations agreed with or similar to the facts, while RAPD markers seemed to fail in revealing the heterozygosity in some populations.The result of UPGMA cluster analysis showed that 15 RAPD markers could reflect the known relationships and genetic variations between any two populations.
