几种鹤性别的分子生物学鉴定
SEX IDENTIFICATION IN SEVERAL CRANES WITH MOLECULAR BIOLGY METHODS
【摘要】 以RAAV0 1和RAAV0 2两条随机核苷酸加聚体为引物 ,通过随机扩增片段多态DNA的方法 ,在白头鹤、白枕鹤、丹顶鹤等 3种 4对不同个体中 ,发现一条约 30 0bp的雌性个体特异带 ,并应用这一方法成功地鉴别了未知性别的丹顶鹤个体 .同时参照已知动物CHD基因序列设计合成CHD基因引物 ,采用PCR方法在丹顶鹤雌性个体中扩增出一条 2 0 6bp的片段 .序列分析表明 ,该序列与已知其他鸟类CHD W ,CHD Z基因的编码区和内含子区都有较高的同源性
【Abstract】 A femalespecific band about 300bp has been found in several cranes with two 10 mer primers RAAV01 & RAAV02 by RAPD. The authors have successfully identified sex of some individuals of red crowed crane with the same method. Meanwhile, part of CHD gene about 206bp has been amplified in female red-crowed-cranes with two 20mer primers using PCR. The DNA homology between the part of CHD gene we amplified and the published sequences of the intron and the exon of CHDW & CHDZ in other species is over 90%.
