瞬态电磁脉冲对细胞增殖的影响

【摘要】 实验采用有丝分裂指数测定法及3H TdR掺入法 ,研究瞬态电磁脉冲对人外周血淋巴细胞及艾氏腹水癌细胞增殖的影响 .结果发现 ,没有植物凝集素PHA时 ,10 0kHz和 2 0 0kHz瞬态电磁脉冲对淋巴细胞有丝分裂指数和3H TdR掺入量无显著影响 ;有PHA时 ,10 0kHz瞬态电磁脉冲促进淋巴细胞摄取3H TdR ,细胞有丝分裂指数增加 ,2 0 0kHz瞬态电磁脉冲抑制淋巴细胞摄取3H TdR ,细胞有丝分裂指数下降 .这两种频率的瞬态电磁脉冲都能明显地促进艾氏腹水癌细胞对3H TdR的摄取 .结果表明 ,瞬态电磁脉冲对细胞增殖是否产生影响 ,依赖于照射时细胞是否处于增殖活化状态 ;这种影响存在频率“窗效应” ,其影响很可能是对胞内增殖信号传递的作用 .

【Abstract】 The effects of transient electromagnetic pulse (TEMP) on the proliferation of human peripheral lymphocytes and Ehrlich ascites cells were studied, by analyzing the mitotic index and the amount of 3 H TdR incorporation. It was found that 100kHz and 200kHz TEMP didn’t produce any effect on the human peripheral lymphocytes mitotic index and 3 H TdR uptake if there was no PHA stimulation during TEMP exposure. However, if with PHA stimulation, the lymphocytes mitotic index and the amount of 3 H TdR incorporation increased by 100kHz TEMP, and inhibited by 200kHz TEMP, compared with control group respectively. Ehrlich ascites cells were exposed to the same two types of TEMP for 0.5~3.0h and the enhancement of 3 H TdR incorporation was observed in both irradiation groups. All these results showed that TEMP did affect cell proliferation rate, and such effect depended on whether the cells were in proliferative status or not. It was clear from the data here that not only physical “window effects” related to the physical parameters used in the exposure system but also biological “window effects” because of the different cell metabolic status were existed. It was also discussed the effects of TEMP on cell proliferation occurred at which cellular level.
