中国人群中微卫星位点DXYS156的多态研究
A STUDY ON POLYMORPHISM OF MICROSATELLITE LOCUS DXYS156 IN CHINESE POPULATIONS
【摘要】 以中国 2个汉族群体和 8个少数民族群体的 52 0名个体为研究对象 ,采用 PCR扩增后聚丙烯酰胺凝胶电泳分离的方法 ,分析了 Y染色体上 DXYS1 56Y和 X染色体上 DXYS1 56X这两个微卫星位点的遗传多态性。结果表明 :在所研究的 1 0个中国人群中 ,共观察到 1 0个不同长度片段的等位基因 ,在 X染色体上的 5个等位基因是 :1 30 bp、 1 35bp、 1 4 0 bp、 1 4 5bp、 1 50 bp,在 Y染色体上的五个等位基因是 :1 60 bp、 1 65bp、 1 70 bp、 1 75bp、 1 80 bp。 DXYS1 56Y位点的多态信息量较多 ,以 1 65bp为 Y染色体上的主要等位基因 ,频率范围在 0 .2 3— 0 .77;等位基因1 60 bp、 1 70 bp和 1 75bp次之 ,频率范围分别为 0 .0 2— 0 .36、 0 .0 1— 0 .50和 0 .0 3— 0 .31 ;而等位基因 1 80 bp频率较低 ,频率范围为 0 .0 2— 0 .0 5。 DXYS1 56X位点的多态信息量较少 ,以等位基因 1 4 0 bp为各个群体中最普遍的类型 ,频率范围为 0 .86— 0 .98,其余类型均较低。DXYS1 56X的基因频率及中国人群中 DXYS1 56Y的基因频率至今在文献中尚未见报道
【Abstract】 The genetic polymorphisms of microsatellite loci,DXYS156Y on the Y chromosome and DXYS156X on the X chromosome were studied by using PCR method followed by PAGE,in 520 males of 10 populations (2 Han subpopulations and 8 ethnic minorities) in China.The result shows that the ten alleles with different length fragments were found in Chinese populations surveyed.Among them,five are alleles 130bp,135bp,140bp,145bp,150bp on the X chromosome,and the others are alleles 160bp,165bp,170bp,175bp,180bp on the Y chromosome.The locus DXYS156Y has a high polymorphic information content,and the allele 165bp with the frequency range from 0 23—0 77 is predominant in Chinese populations,followed by alleles 160bp,170bp,and 175bp with the frequency range from 0 02—0 36,0 01—0 50 and 0 03—0 31,respectively,while the allele 180bp takes the last place with the frequency range from 0 02—0 05.The DXYS156X has a low information content,and the allele 140bp,with the frequency range from 0 86—0 98,is the most common type,while the other alleles have a quite low frequencies.The allele frequency of DXYS156Y in Chinese populations and that of DXYS156X in human populations have not been reported so far.
